What is GHRP-2?

GHRP-2 (Growth Hormone Releasing Peptide-2) is one of the most potent stimulators of endogenous growth hormone secretion currently known. It is an oligopeptide composed of six amino acids, capable of rapidly crossing the oral mucosa and reaching the bloodstream, making it effective even when administered sublingually or buccally.

Numerous studies have shown that GHRP-2 acts directly on the pituitary gland, inducing an increase in GH secretion of up to 7–15 times the baseline levels. In terms of potency, it is often compared to exogenous growth hormone, but with the advantage of stimulating the body's natural production.

Thanks to its low molecular weight, GHRP-2 has high bioavailability and good tolerability. Clinical studies published as early as the 1990s confirmed its effectiveness in children, adults, and the elderly, making it one of the most studied and widely used peptides in the medical and sports fields.

GHRP Group

The term GHRP refers to Growth Hormone Releasing Peptides, a class of ghrelin receptor agonist peptides. Activation of these receptors (GHS-R1a), located in the hypothalamus and pituitary gland, leads to a pulsatile increase in growth hormone secretion.

Initially developed as a more affordable alternative to GH therapy in patients with somatotropin deficiency, GHRPs have since demonstrated much broader potential, particularly in improving body composition, tissue repair, and health.

Mechanism of action of GHRP-2

GHRP-2 is a mimic of ghrelin, the hormone produced by the stomach during fasting. By binding to GHS-R1a receptors, it stimulates the release of GH in a physiological and pulsatile manner.

The increase in GH leads secondarily to an increase in IGF-1 levels, a key protein in the processes of:

  • muscle growth,

  • protein synthesis,

  • strengthening of tendons and bones,

  • immune support,

  • neuronal protection.

Unlike exogenous growth hormone, GHRP-2 works "upstream," exploiting the body's natural mechanisms.

Effects of increased GH and IGF-1

The increase in GH induced by GHRP-2 has marked effects on body composition:

  • Increase in lean body mass

  • Stimulation of lipolysis and use of fats as an energy source

  • Improved muscle and tendon recovery

  • Strengthening bone structure

  • Support for collagen synthesis

Meta-analyses conducted on healthy athletes have shown an average increase in lean body mass of approximately 1.8 kg with elevated GH levels.

Cardioprotective and cytoprotective properties

In addition to the effects mediated by GH and IGF-1, GHRP-2 exhibits independent properties related to its ability to bind to the CD36 receptor. Studies in animal models indicate:

  • improvement in cardiac function,

  • reduction of cardiac fibrosis,

  • protection of cardiomyocytes during ischemic episodes,

  • anti-inflammatory and antioxidant effects.

At the cellular level, GHRPs protect tissues from oxidative stress and hypoxia, promoting cell survival and regeneration.

Side effects

Despite its high efficacy profile, GHRP-2 may have some dose-dependent side effects:

  • increased appetite (due to activation of ghrelin receptors),

  • increase in cortisol levels,

  • increased prolactin,

  • water retention,

  • transient hypoglycemia.

For this reason, its use should be conscious and monitored.

Benefits and limitations of GHRP-2

Key benefits

  • Strong stimulation of growth hormone

  • Increase in muscle mass

  • Improved body composition

  • Strengthening bones and tendons

  • Cardioprotective and cytoprotective properties

Possible side effects

  • Increase in cortisol and prolactin

  • Accentuated hunger

  • Water retention
